ELISA 酶結合物的制備:免疫酶技術(immunoenzymatic technique)又稱酶免疫測定法(enzyme immunoassay,EIA),是繼免疫熒光技術和放射免疫技術之后發(fā)展起來的又一種免疫標記技術。它是把抗原抗體的特異性反應和酶的高效催化作用相結合而建立的。該技術通過化學方法將酶與抗體或抗抗體結合,形成酶標記物,這些酶標記物仍保持免疫學活性和酶的活性,然后將它與相應的抗體或抗原起反應,形成酶標記復合物,結合在免疫復合物上的酶,在遇到相應的底物時,則催化底物產生水解、氧化或還原等反應,而生成可溶性或不溶性有色物質。然后根據顯色的深淺來反映待測樣品中抗原或抗體的含量,如生成的有色物質為可溶性,則可用肉眼比色或酶標測定儀測定光密度值(OD)定性或定量;如生成的有色物質為不溶性沉淀物,同時又是電子致密物質,則可用光學顯微鏡或電子顯微鏡識別和定位。因此,免疫酶技術是一項定位、定性和定量(敏感度可達每毫升ng~pg水平)的綜合技術。常用的酶是辣根過氧化物酶(horseradish peroxidase,HRP)和堿性磷酸酶(alkaline phosphatase,AKP)。
